PBS was then discarded and the pellet resuspended in 24 ml lysis buffer (50 mM Tris, 1 M NaCl, 10 mM MgCL2) supplemented with 0,001% Pluronic F-68: (Invitrogen #24040032), 1300U Salt Active Nuclease (SAN) (Sarstedt #83.1803) and 100 × HALT Protease Inhibitor Cocktail, (EDTA-free Thermo scientific #78439). Cell suspension was then subjected to three freeze / thaw cycles in liquid nitrogen and a 37 °C water bath, respectively. To assure that the suspension does not contain any remaining plasmid DNA, it was again supplemented with 1300U of SAN afterwards, and incubated at 37 °C for 1 h, while shaking at 150 rpm.
